Questions I'm looking at:
- How is transdermal absorption of daro? The prostate cancer research I've found is all about oral.
- To what extent will the metabolism of daro depend on the vehicle? Could significantly affect the dosage calculations and cost?
- Earlier in the thread, Kevand mentioned that daro is a prodrug. Other antiandrogens I've looked at are prodrugs, but I didn't see anything about daro.

Darolutamide is not completely a prodrug. It's a very powerful antiandrogen itself. The active metabolite of it just happens to be a powerful antiandrogen as well, so you can think of it as having two stages of activity. One as it is, and another after one stage of degradation. Both stages are effective.

As far as I know skin cells don't have the enzymes needed to degrade it so degradation will occur via the liver. But I can't say that with 100% certainty.

I’d definitely do it but the cost is the only problem. I imagine over time it will get cheaper when the U.S prescription gets released to the market and more Chinese chemical suppliers start manufacturing it.

Interestingly, the Xi'an supplier emailed me today to say that they're out of stock now of daro. They had 1g left and just sold it to an individual buyer in the US for $430. Normal minimum order size is 5g. I believe them about this and don't think they're just trying to do any high-pressue sales on me; they just let me know about the sale because in case I want to buy, their time to delivery has increased and they would now need 15-20 days to produce from time of order.

Daro market is heating up ;-)

Done. Looks like around 0.1% darolutamide will be enough to clearly beat 5% RU:

Using a higher dose like 0.2-1% should give massive androgenic blockade. Even at 1% and 2.5 mL twice daily, this only means your body is exposed to 50 mg a day. The castration dose (prostate cancer) is 1200 mg per day. So that's 4% of the castration dose you'd be exposed to if everything absorbs through your skin, and that's using a massive strength and volume.

No one should ever need to go higher than 1% if it holds up to its promise.

But that said, once it becomes cheap, mixing a 5-10% topical darolutamide would be so much ridiculously stronger than any RU58841 solution even possibly could be, high strength darolutamide would probably become our first viable "topical nuke".

Keep it up, can't wait for the first results!!

I do have a question: 0,1% DARO is a bit stronger than 5% RU. But if you wan't to nuke it, shouldn't you go higher then 0,1%?
For example; a lot of members tried RU at 5% or 10% but didn't respond to it.

I have no knowledge of chemicals, but in theory, does DARO have a different mechanism so that 0,1% DARO might work, while 10% RU doesn't?

This is a extremely potent antiandrogen.
It will definitely have the potential to have feminizing effects.
Especially when using DMSO as a vehicle...
Something will always go systemic, the question is how much.
So far, we can only do basic calculations.
Then again, oral dutasteride has a profound influence in the body with 100% systemic effects.
You can decide which risks you want to take and which one is riskier than the other I guess?

Shipped via courier? I guess you don't have to worry about getting it intercepted by customs. Lucky you, wherever you live.

Here, everything that gets shipped by courier must have a customs declaration and gets inspected thoroughly. With regular mail, you have a 30-40% chance of getting it without inspection, but it takes weeks.

I guess I will just have to wait for a group buy so someone can reship it. I am following your experiment closely though, PLEASE keep us updated. I'm in deep sh*t with RU. It's not doing much for me anymore. I need the 'next big thing', and if this isn't it, then I don't think anything will be.

Sorry, I don't mean to discourage you - I am just telling you what my main problem is from my perspective.

Do you think I wouldn't already be using enzalutamide and possibly even this if I could reliably order them without a huge risk of losing money and getting into legal issues? I would, since months ago. But I can only wait for someone within the EU to organise a group buy, otherwise no way.
