Two studies [26, 27] using transgenic mice with overexpression of COX-2 in the skin showed directly contradictory results. Transgenic mice under the control of a bovine keratin 5 (K5) promoter[26] and human keratin 14 promoter[27] and overexpressing COX-2 in both the basal cells of the interfollicular epidermis and the pilosebaceous unit, exhibited reduced hair-follicle density and delayed postnatal hair follicle morphogenesis compared with wild-type animals. Administration of a specific COX-2 inhibitor (in one experiment [27]) restored hair growth, indicating that the alopecia was attributable to elevated COX-2 enzymatic activity.

are you serious?? Dude no. Inhibit COX-2 may be easier but by doing that you would also be inhibiting GOOD PROSTAGLADINS. FACT. Why would you want to do that??

restored?? really?? 100% of original hair? That wouldn't make any sense as you would also be inhibiting GOOD PROSTAGLANDINS.
Inhibiting COX-2 MAY work to some extent but you would get MUCH better results if you were to just block PGD2 directly.

Everything is out of balance. this is why PGD2 IS OVER-expressed. Cox-2 has an important regulatory role of the prostaglandins..

Look at this simple study on fish oil..

Objectives

We examined the effect of EPA on prostaglandin (PG) D2 generation in the cultured human mast cells with IgE-anti-IgE challenge incubation.
Methods

Cultured human mast cells were incubated with EPA (1 ?mol/L) for 20 h, then challenged with anti-IgE incubation after treatment with IgE. At the same time, COX inhibitors were tested to identify COX-1 and COX-2 activity. PGD2 synthetic activity was also assayed in a cell-free homogenate of cultured mast cells with COX inhibitors and EPA. Histamine in the culture medium and in cells was assayed with the HPLC-fluorescent method. PGD2 and PGD3 were assayed with gas chromatography-mass spectrometry and the stable isotope dilution method.
Results

Although EPA incubation did not affect histamine release by cultured human mast cells in response to IgE-anti-IgE challenge incubation, it did decrease PGD2 generation by inhibiting the COX-2 pathway. In contrast, in the cell-free homogenate of cultured human mast cells, EPA inhibited both COX-1 and COX-2 activities.
Conclusion

Pre-incubation with EPA primarily affects the COX-2 pathway in cultured human mast cells and reduces PGD2 generation in response to IgE-anti-IgE challenge incubation. These findings suggest that COX-1 and COX-2 have different substrate flow systems in mast cells. They also suggest that endogenous EPA diet supplementation would reduce PGD2 production and could serve as an anti-inflammatory substrate in human mast cells.

The guy overdosed on Sulfasalzine.. Sulfasalazine inhibits nuclear NF?B pathway so it reduces Cox-2 and prevents it to elevate again..

^ not sure if I understand, but where does it say that it does not affect other prostaglandins such as PGE2 and PGF2...

if COX-2 was the root of the problem, then wouldn't have cotsarelis study picked up on that?

Thyme Oil Can Inhibit COX2 and Suppress Inflammation

When Inoue and colleagues analyzed thyme oil, they found that the major component -carvacrol- was the primary active agent; in fact when they use pure carvacrol extracts in their tests COX-2 levels decreased by over 80%.

Abstract

Cyclooxygenase-2 (COX-2), the rate-limiting enzyme in prostaglandin biosynthesis, plays a key role in inflammation and circulatory homeostasis. Peroxisome proliferator-activated receptors (PPARs) are ligand-dependent transcription factors belonging to the nuclear receptor superfamily and are involved in the control of COX-2 expression, and vice versa. Here, we show that COX-2 promoter activity was suppressed by essential oils derived from thyme, clove, rose, eucalyptus, fennel, and bergamot in cell-based transfection assays using bovine arterial endothelial cells. Moreover, from thyme oil, we identified carvacrol as a major component of the suppressor of COX-2 expression and an activator of PPARalpha and gamma. PPARgamma-dependent suppression of COX-2 promoter activity was observed in response to carvacrol treatment. In human macrophage-like U937 cells, carvacrol suppressed lipopolysaccharide-induced COX-2 mRNA and protein expression, suggesting that carvacrol regulates COX-2 expression through its agonistic effect on PPARgamma. These results may be important in understanding the antiinflammatory and antilifestyle-related disease properties of carvacrol.

i have an interesting experience about hairloss and getting cold in past two years and maybe it is related to this thread. In past two years whenever i've got cold i've used adolt cold tablet and licophar and surprisingly during the treatment my hailoss decreased very much! I think there is a relation between adolt cold tablet and hairloss.

Is it this simple:

PGE2 and PGF2 stimulates hair follicle growth.
PGD2 inhibits hair growth.

^ When all of that is in balance, you end up with a full head of hair. That extra PGD2 on non-balding people is only there to keep those "stimulants" in check so you don't end up like this guy:

I think if you were to maximize those GOOD PROSTAGLANDINS and completely eliminate PGD2, then you could probably turn any NW7 guy into the guy pictured above :punk: :punk: :punk:

PGD2 excess is the side-effect of over-expressed COX-2. COX-2 is a inflammation mediator , catalyzes the conversion of the free essential fatty acids to prostanoids a sub-class of Eicosanoids. Prostanoids mediate local symptoms of inflammation: vasoconstriction or vasodilation, coagulation, pain and fever. Inhibition of cyclooxygenase, specifically the inducible COX-2 isoform, is the hallmark of NSAIDs (non-steroidal anti-inflammatory drugs), such as aspirin. COX-2 is responsible for pain and inflammation, while COX-1 is responsible for platelet clotting actions.

Prostanoids activate the PPAR? members of the steroid/thyroid family of nuclear hormone receptors, directly influencing gene transcription.

Is Cotsarelis is going to release anything soon? :)

If PGD2 is the real culprit of hair loss.. Why nothing has been done yet? :woot: It is just a billion $$$$$$ industry.
