my dutasteride pills are liquid, they are on soft capsulesHello guys, I have some updates about my dutasteride solution. I've been reading lot of studies and it seems mct + ethanol alone causes good solubility on dutasteride but the bioavailability remains less than 20% because the ethanol is rapidly absorbed by the body. After that happens most of the dutasteride precipitate from the solution because dutasteride is not very soluble in mct oil. For this reason I have been reading lots of studies and I will incorporate SEDDS(Self-emulsifying drug delivery system) with this method I will basically be able to form micelles with surfactants that encapsulate the dutasteride in the oil preventing separation of it and increasing bioavailability. I will use a ratio of 43.59% Smix (Smix is surfactants + cosurfactants) which should decrease droplet size to <100 nm as per studies. Basically when you have <100 nm droplets the drug is highly bioavailable and it can absorb pretty good. Also when it reach the small intestine and make contact with bile salts, these can drop the dropplet size to like 30 nm droplet size which would absorb the drug pretty damn easy. With this method i will achieve a bioavailability very similar to Avodart because the solution wont precipitate(cause I wont rely on ethanol to dissolve dutasteride, the solvents of dutasteride will be lecithin and polysorbate 80 which have high solubilty in dutasteride, you can check it here https://pmc.ncbi.nlm.nih.gov/articles/PMC4232777/#:~:text=of drug without addition of,Figure 2). I will mostly use natural surfactants because surfactants at high amounts are hazardous, the only synthetic one I will use is Tween 80 but in small dosages which is under the ADI(Acceptable Daily Intake) of 25 mg/kg(I will use ~3.8mg/kg).

They use SEDDS as well. The capsule contains lecithin that is a natural emulsifier and it also contains mono and diglycerides of capril/capric acid which solubilizes the dutasteride good and is another emulsifier. The pill is fast dissolved in the stomach and the lecithin on the pill + the mono and diglycerides start to emulsify the dutasteride in presence of the stomach acid which is mostly made of water. When emulsifiers/surfactants get in contact with water, they make micelles which traps the lipophilic drugs, that in other cases it would be non bioavailable to be used by the body(the body cant metabolize dutasteride if it is not encapsulated with micelles, that is basically why taking the raw powder with water wont work and that is also why taking only oil with it wont work because it will separate from the oil in the stomach in the absence of an emulsifier) . In my case I use lecithin like avodart but also a strong synthetic surfactant which is tween 80 because dutasteride has trash solubility in mct oil and getting mono and diglycerides is basically impossible to get if your not a chemist or pharmacist, or it is too expensive, so I have to rely in very potent synthetic surfactant such as tween 80 to solubilize most of the dutasteride with the help on lecithin ofc. I also use some monolaurin to get some stability too and ethanol to make the dissolution faster outside the body and to help reach smaller droplet size(which translate to more bioavailability).my dutasteride pills are liquid, they are on soft capsules

So i currently prepare a little experiment on DIY oral dutasteride solution and want to share it with you guys:
We all know 2.5mg dutasteride is expensive so why not create our own 2.5mg dutasteride. I have ordered raw Dutasteride powder from a verified source which will arrive soon.
I will create a solution with Dutasteride powder, MCT Oil and Vitamin E that has 2.5mg of dutasteride per one ML. I have a 0,001mg scale on hand and all my laboratory equipment
I did some research and the whole Dutasteride capsule contains: An oil (very similar to MCT, Dutasteride itself, an anti-oxidant and a gelatine capsule.
With my solution i want to recreate this without the gelatine capsule:
The solution would be stored in a cool place, with a bottle that blocks UV or any Light.
Why MCT Oil: Well, dutasteride is lipophilic and its basically in the original capsule
Why Vitamin E: It is an anti-oxidant and prevents degradation. Usually, it is better to use BHT-Powder to have the best stability. But i will make 3 months batches and after discussing that topic with my Professor in Uni, i wouldn’t actually need an antioxidant.
But to be extra safe i will use Vitamin E and only make solutions for 3 months.
I will also follow some other tricks, like using a vial and getting the solution out with a syringe. But that would be a too long post
So i am 100% sure that i will have nearly pharma level degradation protection
5 grams of powder and all the other stuff costed me around 140€. And the powder would last years
So every evening i will open the bottle and drink 1ml of it.
